Abstract :
Kurkumin diklasifikasikan ke dalam BCS (Biopharmaceutical Classification System) kelas IV berdasarkan kelarutan dalam air yang buruk (4,375 ?g/mL) dan bioavailabilitas yang rendah (1.498 ± 0.402 ?g jam/mL) (Shelma & Sharma, 2013). Oleh karena itu, diperlukan teknik untuk meningkatkan bioavailabilitas dalam kurkumin salah satunya yaitu nanopartikel. Penelitian ini dilakukan untuk mengetahui inhibisi dan nilai IC50 terhadap aktivitas penghambatan enzim ?-glukosidase secara in vitro. Hasil dari nanopartikel kurkumin diuji aktivitas inhibisi terhadap enzim ?-glukosidase menggunakan spektrofotometer Uv-Visible pada ?=305 nm dan dihitung persen (%) inhibisi serta nilai IC50 nya. Uji aktivitas nanokurkumin terhadap penghambatan enzim ?-glukosidase secara in vitro menunjukkan nilai IC50 3,95 ?g/mL sedangkan akarbosa menunjukkan nilai IC50 akarbosa yaitu 4,11 ?g/mL. Aktivitas inhibisi nanokurkumin terhadap enzim ?-glukosidase menunjukan nilai IC50 yang lebih baik dibandingkan dengan akarbosa dan termasuk ke dalam kategori sangat aktif. Kata kunci: kurkumin, enzim ?-glukosidase, nanoteknologi. Curcumin is classified into BCS (Biopharmaceutical Classification System) class IV based on its poor water solubility (4.375 g/mL) and low bioavailability (1,498 ± 0.402 g hour/mL) (Shelma & Sharma, 2013). Therefore, a technique is needed to increase the bioavailability of curcumin, one of which is nanoparticles. This research was conducted to determine the inhibition and IC50 of the inhibitory activity of the ?-glucosidase enzyme in vitro. The results of the curcumin nanoparticles were tested for the inhibitory activity of the ?-glucosidase enzyme using a Uv-Visible at ? =305 nm and the percent (%) inhibition and IC50 value were. Against the inhibition of ?-glucosidase enzyme vitro showed an nanocurcumin 3,95 ?g/mL while acarbose showed an IC50 value of in acarbose that was 4,11 ?g/mL. Against the inhibition ?-glucosidase enzyme showed nanocurcumin a better IC50 value than acarbose, and included in the very active category.
