Abstract :
Aspartate Aminotransferase (AST) is an intracellular enzyme that
is very important for the body, so the result of the examination must be released
quickly. One that can be done is to shorten the time and increase the speed od
centrifugation. This research conducted centrifugation at 5000 rpm for 1 minute.
Examination using sample in the form of serum.
Objective : To determine the diffrence in serum AST levels from blood centrifuged
at 3000 rpm for minute and 5000 rpm for 1 minute.
Methods : This type of research is Pre-experimental with a static group
comparison research design. The subjects of this study were 32 respondents who
had met the inclusion and exclusion criteria. The blood samples were then frozen
at room temperature for 20 ? 30 minute, then centrifuged at 3000 rpm for 5
minute and 5000 rpm for 1 minute. The examination method used is the IFCC
method without pyridoxal. The data were analyzed using Wilcoxon Signed Rank
Test statistical test.
Result : Aspartate Aminotransferase (AST) levels in blood serum that were still
within norma limits. The normality test shows that the data is normally
distributed, so to test the hypothesis using a Paired T test. The result of the there
is no significant difference in serum from blood centrifuged at 3000 rpm for 5
minute and 5000 rpm for 1 minute wiht a pvalue (Sig2-tailed) 0f 0,262 > 0,05.
Conclusion : Based on the resultsof this study, it can be conclude that there is no
diffrence in the level of Aspartate Aminotransferase (AST) in serum from blood
centrifuged at 3000 rpm for 5 minute and 5000 rpm for 1 minute.
Suggestion : Applicatively the management of examination material, namely the
centrifugation speed of 5000 rpm for 1 minute, can be an alternative of
consideration in the AST examination.
