Abstract :
The purpose of this research was to know the activity of antibacterial test
of ethanol extract of jukut pahit leaves (Paspalum conjugatum) of Pongruan
village of East Manggarai regency towards to the growth of Staphylococus aureus
bacteria and the study of its active compund. The total of samples of jukut pahit
macerated leaves were 200 g with 70% ethanol solvent, and the obtain of viscous
extract is 29,90 g. The result of rendemen was 14.95%. The physical properties of
the extract are: blackish brown, sticky and soluble in cold water. Phytochemical
test results are: positive contains alkaloid compounds, steroids, and Saponin. The
results of antibacterial activity test of Staphylococcus aureus bacteria with
diffusion well method obtained by the data as follows 75% and 100%
concentration have very strong inhibitory power, concentration of 1,45%, 2,25%,
5,25%, 10,25%, 12,5%, 16,25%, 25%, and 50% has a strong inhibition.
Fractionation of the active compound in the sample with TLC and column
chromatography using eluent ethyl acetate: chloroform (1:1 v / v) obtained 40
fractions, grouped into 6 groups of fractions based on Rf value and spot equality.
Analysis of the active compound on the sample fraction using UV-Vis and
infrared spectrophotometer, it was found that fraction A denotes absorption bands
of λ 230 nm. Fraction B has an absorption of λ 225 nm. The C fraction has an
absorption of ï¬ 220 nm. Fraction D has an absorption of ï¬ 225 nm. Fraction E has
an absorption of ï¬ 230 nm. The F fraction has an absorption of ï¬ 230 nm which
shows the electron transitions ππ*. The infrared spectra of fractions A, B, C, D,
E and F indicate the presence of an alpha, N-H, C-H alifatik, C-H aromatik, C-N,
C=N, C=C dan C-O.
