The Effectivity Of 2IP And IAA Giving Through Black Potato

ABSTRACT
Black potato (Plectranthus rotundifolius) is one kind of plant tubes which important as a source of carbohydrates. This plant is one of the staple foods in several countries, especially Asia and Africa, because it contributes the largest calories in the main food once consumed. Potato cultivation is generally done by planting the tubers. Cultivation using this method continuously can result in a tendency to decrease yields in the next generation. Therefore an alternative propagation by tissue culture is needed. The purpose of this research was to determine the correct concentration of ZPT 2-iP and IAA for the multiplication of black potato shoots. The research was carried out from March 2020 to August 2020 at the Jember State Polytechnic Network Culture Laboratory. This research used a factorial Completely Randomized Design (CRD) with 2 factors, 9 treatment combinations, and 3 replications. The first factor, namely ZPT 2-iP consists of 3 levels, namely 1; 3; and 5 mg / l. The second factor is ZPT IAA consisting of 3 levels, namely 0.25; 0.50; and 0.75 mg / l. The results showed that the optimal concentration of 2-iP was added to the stem diameter of black potato plants in vitro, where the optimal concentration for stem diameter parameters was 1 mg / l. The addition of IAA concentration significantly affected the treatment combination. Treatment with 2-iP concentration of 3 mg / l and IAA of 0.75 mg / l was optimal for the number of leaves and plant height parameters, 2-iP 5 mg / l and IAA 0.75 mg / l was optimal for the number of shoots, 2-iP 1 mg / l and IAA 0.75 mg / l were optimal for root length, and 2-iP 1 mg / l and IAA 0.25 mg / l were optimal for the number of roots.
